Abstract The lipase from Candida rugosa was coated with synthetic didodecanyl glutamate ribitol and used to catalyze esterification of lauric acid and lauryl alcohol in organic solvents. The optimal reaction conditions were determined, which are buffer pH for lipase coating,6 8;reaction temperature,30℃;solvent,isooctane.The kinetics of the esterification reaction obeyed the Ping-Pang Bi-Bi mechanism with an inhibition by lauryl alcohol. The kinetic parameters under the optimal conditions were determined. The Michaelis constants of lauric acid and lauryl alcohol were 29 1 and 71 4mmol·L -1 ,respectively. The maximum rate was 0 66mmol·min -1 ·g -1 .The inhibition constant of lauryl alcohol was 11 3mmol·L -1 .The results predicted by the kinetic model were in good agreement with experiment.
