Abstract The product of recombinant human interleukin-6 was expressed and existed in the form of intracellular protein inclusion body in the cytoplasm of recombinant E.Coli cell. A systematic study was carried out on the influence of physical and chemical factors including power output,disrupting time,cell concentraction,buffer pH,cell frozen and suspension system during ultrasonic process.The conditions of disruption and release of inclusion body was determined:by ultrasonic ouput power 480W,cell concentration 100g/L,in the deionized water suspension system,obtained 40% original inclusion body from the biomass with 20% expression level.
