Abstract The response surface methodology (RSM) was used to obtain optimal medium components for Neosartorya fischeri G5 β-mannanase production. 8 factors were selected for Plackett-Burman design based on single factor experiment to evaluate the influence. The steepest ascent experiment and central composite design were adopted for further optimization. Design-Expert statistic methods were used for quadratic regression model equation to find out the optimum concentration levels and the relationships among factors. The optimal medium consisted of guar gum 13.0 g/L, peptone 11.0 g/L, yeast powder 5.0 g/L, MgSO4 1.0 g/L, KH2PO4 0.5 g/L, NaCl 0.6 g/L and CaCl2 0.6 g/L at pH 4.0, and the final enzyme yield was more than twice of the original.
