Preparation of Hydroxypropyl-β-Cyclodextrin and Its Solubility Improvement on Dexamethasone

Abstract The inclusion behavior and solubility improvement on dexamethasone of 2-hydroxypropyl-β-cyclodextrin and β-cyclodextrin in aqueous solution were discussed.Experimental results showed that 2-hydroxypropyl-β-cyclodextrin with different substitute degree were all able to increase the solubility of dexamethasone,2-hydroxypropyl-β-cyclodextrin had stronger ability than β-cyclodextrin.The solubilization effect of 2-hydroxypropyl-β-cyclodextrin which was prepared in 0.5%,1.5% and 30% NaOH solution increased with the increasing of substitute degree of 2-hydroxypropyl-β-cyclodextrin,but the solubilization effect of 2-hydroxypropyl-β-cyclodextrin which was prepared in 3% NaOH solution decreased with the increasing of substitute degree of 2-hydroxypropyl-β-cyclodextrin,and this might have some relation with the different substitute position of 2-hydroxypropyl-β-cyclodextrin.At the same time,the solid inclusion complex of dexamethasone with β-cyclodextrin was prepared through cosolvent method,and the structure of the solid inclusion complex was studied.
