Abstract Anaerobic dark fermentation experiments were performed in microcosms to produce biohydrogen from glucose. The influences of pH, concentration of NH+4, Fe2+ and Mg2+, and the temperature and duration of preheating on the hydrogen production efficiency were evaluated via single factor experiments. The optimal conditions were concluded to be preheating for 40 min at 90 ℃ followed by dark fermentation at pH 8.0 with 3.00, 0.10 and 0.03 g·L-1 of (NH4)2SO4, FeSO4 and MgSO4, respectively. Under these conditions, 630 mL high quality H2 (purity: 0.80) was harvested cumulatively with a yield of 2.62 mol H2 per mol glucose. The overall results suggest that anaerobic dark fermentation is an effective approach for biohydrogen generation.
