Abstract Based on the single factor experiment and the orthogonal design, taking the retention rate of polygonapolysaccharose and impurity removal rate as markers, the flocculation process by chitosan in treating the extracted polygonatum decoction was investigated. Experiment factors and levels were firstly selected by one factor tests. The four factors, concentration of waterextraction, chitosan dosage, pH value and flocculating temperature were chosen for the orthogonal design. The results showed the optimal conditions were: the concentration of waterextraction was 4∶10(g:mL), the concentration of chitosan was 15% , the pH value was 4.0 , the flocculating temperature was 40 ℃ and the time was 3 h. Compared with alcohol precipitation method, by chitosan flocculating method, the flocculation rate was 16.08% and 49.75%; the retention rate of polygonapolysaccharose was 88.42% and 60.34%; colligation score was 66.72% and 57.16%, respectively. The flocculation of polygonatum L. extracted with chitosan is effective and it has greater advantage than the ethanol precipitation method. |
