Abstract Mutual combination mechanism of DNAF with bovine serum albumin (BSA) was investigated by applying watersoluble 4,9di[N(2dimethylamino) ethyl]9aminoacridine4formamide(DNAF) probe. The corresponding thermodynamic parameters ΔH, ΔG and ΔS were calculated and through fluorescence quenching method, the number of binding sites n and apparent binding constant KA were determined. The distance of donor BSA and receptor DNAF, namely r was derived based on the theory of fluorescence resonance energy transfer. The quenching mechanism of DNAF on intrinsic fluorescence quenching of protein and the types of interaction forces were proposed.
