摘要 在摇瓶中对基因重组脑钠素 (BNP)在大肠杆菌中的表达进行了研究 ,确定诱导后最佳培养时间为 4h ,表达量达 2 1 8%。采用金属螯合亲和层析法对十聚体脑钠素进行了纯化 ,比较了咪唑洗脱法和降pH值洗脱法对目标蛋白的纯化效果 ,得到了一条纯化目标蛋白最佳亲和层析纯化路线 ,为后续实验打下了很好的基础

Abstract: Expression of recombinant 10BNP in E.coli was studied in shake flasks and the optimum induction time is 4h.The expression level was over 21 8%.10BNP fusion protein which was produced as inclusion body (IB) was separated by breaking bacteria through sonication.IB was properly dissolved in the presence of 8M Urea.By means of Immobilized metal ion affinity chromatograph(IMAC),comparing the results of different elution methods,a optimum purification route was attained.The work facilitates the further research.
