摘要 应用水溶性4,9二[N(2二甲氨基)乙基]9吖啶胺4甲酰胺(DNAF)探针,研究了DNAF与牛血清白蛋白(BSA)的相互结合作用机理。计算出相应的热力学参数ΔH、ΔG以及ΔS;通过荧光猝灭方法研究结合作用,测定出结合位点数n和表观结合常数KA;根据荧光共振能量转移理论求得供体BSA和受体DNAF的距离r;研究了DNAF对蛋白质内源荧光猝灭的猝灭机制和主要作用力类型。

Abstract: Mutual combination mechanism of DNAF with bovine serum albumin (BSA) was investigated by applying watersoluble 4,9di[N(2dimethylamino) ethyl]9aminoacridine4formamide(DNAF) probe. The corresponding thermodynamic parameters ΔH, ΔG and ΔS were calculated and through fluorescence quenching method, the number of binding sites n and apparent binding constant KA were determined. The distance of donor BSA and receptor DNAF, namely r  was derived based on the theory of fluorescence resonance energy transfer. The quenching mechanism of DNAF on intrinsic fluorescence quenching of protein and the types of interaction forces were proposed.
